Summary/Transcribed: a. Initiated when C1 recognises a microbial surface directly or binds to antibodies already bound to a pathogen. b. Binding causes cross proteolysis and activation of C1r and C1s proteases -> Activated C1 c. Activated C1s cleaves C4 into C4a (acts as anaphylatoxin*) and C4b which covalently attaches to the surface of the pathogen. d. C2 coenzyme binds to C2b and is then cleaved by activated C1 releasing C2a, creating Protease C4bC2b** = C3 convertase e. C3 binds to C3 convertase and is then cleaved by it as well releasing another anaphylatoxin C3a while C3b binds covalently to Pathogen surface and associate with C4bC2b forming C4bC2bC3b = C3/C5 convertase f. Many molecules of C3 can be cleaved by C3/C5 convertase releasing many molecules of C3a and C3b (binds to surface of pathogen inducing phagocytosis). g. C3/C5 convertase also cleaves and activates C5 releasing C5a (potent anaphylatoxin and most important chemoattractant) and C5b which then acts to form the initiation of formation of the membrane attack complex (MAC). h. C5b (not membrane associated) assembles with C6 and C7 (membrane associated) and insert themselves in bacterial membrane i. C8 binds to C5bC6C7 complex and inserts as well into cell membrane forming C5bC6C7C8 complex j. Complex catalyses assembly of many C9 molecules creating cylindrical pore k. Pore disrupts cell balances and kills cell. *Play a role in inflammation, chemotaxis, and the activation of immune cells. **New nomenclature.
