Yes, BUT How did you get pDNA from mRNA? was it by reverse transcription, followed by cloning the cDNA into a plasmid vector, such as pmRNAxp? and then use the plasmid DNA as templete for in vitro transcription? these are not clear from this talk!?

Hi and thank you for your question. As shown on the slide, plasmid DNA is isolated from E. coli, then linearized using a restriction enzyme. Linearized plasmid DNA is used as a template to produce mRNA in IVT reaction. mRNA is then purified from IVT as described in the webinar.
