This was such a useful video and explained things so well but I'm a bit confused as to when the ZFNs were developped because when hes speaking he says 1:20 it was developped in 1994 but on the actual screen it says 1:26 "1985: discovery of zinc finger nuceases". I would love it if someone could pls explain it to me.

Good catch! Zinc finger nucleases were _discovered_ in 1985, but they were first _used_ in the context of genome editing in 1994. It's kind of like how microwaves (the form of energy) were discovered in the 1800s, but they didn't invent the microwave oven until the 1940s.

I actually don't know of any beginner books about CRISPR-Cas9 itself (will update this comment if I find any), but "The Code Breaker" is a wonderful book that goes through the history of the discovery of gene editing technology and is relatively straightforward to understand. If you get the chance to read it, let me know what you think!

Beginning to go into , interfering with natural processes. Not advisable. “ Gain of function” for power of humans for financial gain or weaponise our DNA as they require. Not ours to be messing with means , DNA is a random with many variables . Ability to go there is not advised. But once the ability has been spread, like Guns,. . Tool or weapon, or profit use likely to become abuse.

We have, say 28 billion cells esch with a complete set of DNA. CRISPR is the method to cut out deffective Gens in the DNA of one Cell and replace it with a new Gen. How will thereafter all 28 billion -1 update their DNA? I have no clou as the multitude of humans have. Will appreciate some factual insight. Thanks

Most understandable and clear explanation of CRISPR. This makes me want to take Chem 177! And I had no idea David Liu's lab is doing this cutting edge research...amazing. CCB is so cut off from what the Medical School and Broad labs are doing. :(

These aren't opposites! "N" just means any nucleotide can be in that spot (A,T,C,G), so "NGG" can refer to AGG, TGG, CGG, or GGG.
