Eb is the enharmonic equivalent of d#. Most basic tuners nowadays will give you d# instead of eb, thus the reason why I said d# instead of eb. Likewise a# is the enharmonic equivalent of Bb. Since most tuners nowadays will give you a# instead of Bb, I advised the viewer to tune the 2nd string down to a#. A# is absolutely one semitone below B, just as d# is absolutely one semitone below E. I hope this explanation helps.
