Thank you for the video. It is really clear and helpful. But in our lab, it is very strange that our GC-SCD gives decaying signals time by time. We tried to inject Sulfur standard gas and each time the peak area reduced by 2~3%. But if we let the instrument take a break for one hour, the signal rises in the next shot. Agilent Engineer came and replace the loop, valve and something else, but it has no change. We are still trying to solve the problem.

Thanks. Can we able to predict the no of isomers present in the mixtures using the LCMS/MS

It depends. Some isomers will react differently with HPLC column, and therefore will have different retention times. For example, a branched chain isomer will typically have less interaction with the packing of the column, and will elute early, while a straight chain isomer will tend to have more interaction with the column and will elute later. In cases like this, you will see multiple peaks in a chromatogram, which each represent a different isomer. However, this is very dependent on the type of isomer (for example stereoisomers, or large molecules with isomers that are very similar in structure will be difficult to separate) and also dependent on the relationship between the compound and the column, and the mechanism of interaction between the two. I hope this is helpful.

I currently work for ThermoFisher scientific in the UK but I am studying my Masters degree in environmental assessment and control. This video was helpful in my paper on methods of testing drinking water for fluoride contamination.
