0:02- Aim 1:00 material required 2:00 glacial acetic acid and alcohol in ratio 1:3 Onion bulb 2-3 cm for 1 hr in 1 watch glass ---+ And another watch glass me Hcl is taken --where root tips and the soln is dipped Then we heat - ----for few min We put these root in ----Acetocarmine Then the root are taken on slides then covered with cover slip ------over it and make sure precautions no air bubbles then gently pressed

Mam I have a doubt regarding the water holding capacity that the filter paper also absorbs some of the water that you have poured into the funnel, so how do we know the exact water holding capacity.

I do this experiment at home for practice I used chilled hand sanitizer instead of ethanol. And I isolated the DNA of guava. Tomorrow is my bio practical.

Mam detergent contain proteases, lipases but do not contain RNAase. So the DNA we extracted also contain RNA in it.

Mam practical was superb Mam asked the questions in viva about from quadrat that density application where we can apply this in our surrounding and life mam can you tell it's answer. @@AkashdeepSingh-pv2hi I never tried ethanol at room temperature, as it's also mentioned in ncert that dna extract by adding chilled ethanol. Don't worry, don't be afraid. Everything will be ok👍👍 You can tell your external also if chilled ethanol is not available in your school. All the best👍 Tomorrow Share your experience also.
