I was just clicking video after video looking for someone who skips the soak, skipped it with popcorn and some gypsum powder and having good results so far, didn’t know how different this would be, but I think doing the 30-40 min simmer makes more sense if it works

The simmer gives me the best results, but without the soak I've had contamination pop up more often.

@ it unfortunately ended up getting dumped and I’m starting over with diff projects, but I had loads of agar in there for like 5 days and it didn’t spread at all, I was attempting a soak for the 2nd try and I had to restart, this next week I’m going to soak for 12-18 hours, then boil for like 5-10 (until I see a burst grain, then out and let steam dry for like 45 mins and then in jar and instant pot for like 2 hours, the grain with only simmer might have been working, but there was a lot of burst grains and it didn’t appear the grains actually retained enough water, but maybe this was just too long of air dry time, regardless, I’ll give it another go, for corn, I had no issue at all, 25 min PC in instant pot with gypsum, then dry, jar, hour 40 on high in IP then I had very rapid growth, I was only trying to get the rye down because I heard it’s one of the best, but specifically for growth of sclerotia, or truffles, so I’m going to keep trying, I think it’s a load of fun, I’m obsessed with trying more things and expanding my knowledge on the process. Using a SAB in front of a flow hood should keep things clean.

It's not true laminar flow. But it does work and I haven't had problems with contam using it. It's a cheap way to stay out of a SAB.

Work on cleanliness, a SAB works just as good if you use them right. I have both a SAB and FFU and I'll be honest I use my SAB way more because it provides way more space. FFU's are usually just for commercial growers or growers on a larger scale because it's alot quicker. Alls ik is be super clean and you won't have a problem.

You can try breaking it up and shaking it but when it stalls out I haven't seen them come back to life. I noticed mine stalling when the grain didn't absorb enough water before sterilization.

Remember to work a bit higher as your plates are directly on the desk bu the flow hood blows turbulent air in this area. Just use a wire rack, that should do the trick :)

Remember kids, bleach + rubbing alcohol = chloroform. If your SAB is all bleachy inside, don't be spraying alcohol in there! Also, bleach & water isn't very effective at sanitizing plastics if you're just going to spray and wipe it around. It beads up on the plastic and therefore doesn't provide the necessary contact time. Instead, use a bleach based disinfectant that has the surfactants necessary to wet-out the surfaces you're trying to disinfect. Something like Clorox Bleach Foamer, or Clorox Bathroom with Bleach. And don't be cheap and simply add dish soap to a bleach solution- the two will react and release poisonous chloromine gases.

This is a good point. I use star san which is specifically meant for surface sanitizing using phosphoric acid. When you spray, it doesn't just bead up but clings and spreads on the surface. It is even food safe and good to use in kitchen

Great video, quick note (a bit nit picky but you may find it interesting) you can usually tell the difference between spore prints and transfers because there will be a raised surface(where you dropped your agar)- and likely more Rhizomorphic growth (only the case with transfers, let’s say you use 10 agar for a spore syringe you will have a hell of a time distinguishing them unless only one shows Rhizomorphic growth). Rhizomorphic growth is seen as ideal because it will inoculate spawn faster, however some claim it has the chance to weed out great genetics because the colonies DNA pool shrinks with each transfer. TLDR- Probably not a super useful comment, but if your a mushcult nerd like myself you might find it interesting.

I brought some ready made agar plates online and the para tape didn't come off that easy. Wots the trick coz mine were like glued on

Mine come off really easy and I've never had a very hard time with it. Was the tape flexible or did it tear? Maybe it was scotch tape?

Likely you were sold some very old agar plates, the cohesive material can become “glued” if left on waaaay too long, if this is the case the agar is likely dry and unsuitable for cultures anyhow. Did you ever fix this problem? Or did you just purchase new agar? If you have not seen Philly Golden Teachers Ez as “123” agar recipe give that a view and a shot, agar is not as difficult as people make it out to be and learning it early on is the key to success with this hobby. Mush luck! Edit- If you do PGT 123 Tek DO NOT use Oven Tek for the agar pour, you might get lucky if your in a new house but you will likely have a much higher contamination rate than with SAB.

I've been cultuvating for a little over a year now and have made several hundred plates. I have never used a laminar flow hood and have only ever had one plate contaminated. Proof if you take your time and keep everything clean, you don't really need one.

Agreed! I get a little stressed out working in the SAB because the opportunity for contam seems so much higher than if you're using the flow.

Nice! How many CFM is your pc fan? Is it a 120mm pc fan? Second question, did it works well (i mean, no contaminations)? And last question do you think it is better to use a larger box than your or not ( i mean the space between the filter and the fan) ? Thank you!

This is the fan, amzn.to/3B87mJI its adjustable speed. I put a flame to the front and power it up and adjust the fan speed until the flame is bending but stays lit across the entire surface. Personally, I prefer more space. It works great as long as you don't leave the clean air. With the size being as small as it is, it's easy to let your hand get out of the clean air. I never had any contam show up on the plates I tested. I left the plates uncovered for 2 minutes sitting directly on the table (not up in the air like is recommended for the cleanest air) and I kept them for over 4 months.

Thank you for your risponse, i will probably build one like this, very cool design! I just don't know if i have to let more space between the fan and the Hepa filter or not. I see that lots of flowdood have like 30cm of space beetween the back and the front (filter). I dont know it will maybe creat à more laminar homogenous flow than puting the fan just directly the filter? And also if it's better to put the fan on the top or on the side or on the back? I just don't know... maybe you know? But very Nice design! Looks great!😊 and very cool to hear that it's effective!

this is not a laminar flow hood. This does not provide laminar flow, nor does it have a hood apparatus. Cool little design though, if it works well. Dont need actual laminar airflow, nor the hood to get some good work done. Could possibly help out a lot of people!

I bought "rye berries" and took them to the feed store, the guy showed by whole wheat oats and they looked the same to me. Perhaps I was given bad info?

@MushLoveBeWellFarms Yeah there are a few grains out there, My guess is they got it wrong as a one-off. There are Whole Oats, Millet, Milo, Popcorn Kernels(Food grade), Rye berries, WBS (Wild Bird Seed), and Brown Rice. There's more but those are the main ones used as grain spawn
