Yes, there are buses. You can take bus T10, T13, or BmjP101.

How many hours in total from bangkok to KL? Is it better idea to travel by Train ?Is it safe ? do we need to book a ticket in advance ?

I would say the journey time is more or less around 25 hours. This is not including the waiting/transit time. It also depends on whether you take the express or non-express train on the Malaysian side. From my experience, it's very safe. And yes, you do need to book tickets in advance.
