I know most people say Goku's first transformation as a Super Saiyan is the best, but I actually remember that being rather anticlimactic. Very meaningful, but it happened pretty quick. I'd say the best was Goku's turning into SS3 for the first time, and Gohan going SS2 here was a close 2nd. You could feel the power and emotion in his cries for Cell to stop hurting his family and friends.
