HI, Julien, We have found atleast 10ml of water resudue on head after filteration. Do we need to open the head every time to ensure contamination free operation ?? Second thing is , this entrire operation need further validation as there are many concerns as mentioned bleow, 1) Water residue inside head can lead to bacterial growth and contaminate the Analysis. 2) detaching the membrane from funnel is require more precision compare to usual Filtration method. 3) using the funnel lead as petriplate require validation.

If I receive a RM ATCC derivate with already three passages, does it mean that I could make no more than two passages in the lab? Moreover, from EURACHEM Guide 2023 is written that "Commercial derivatives of reference strains may only be used as working cultures" meaning that, if I buy ATCC derivates, it is suggested that I don't have to make stock cultures?. Hi Davide, According EP and USP it is recommended that “Seed lot culture maintenance techniques (seed-lot systems) are used so that the viable micro-organisms used for inoculation are not more than 5 passages removed from the original master seed-lot.” So that means it is possible to do 2 subcultures. But for example the Vitroids and Lenticule discs are intended for direct use as they are in ready to use concentration or can be directly diluted and show the typical characteristic properties. They are certified for the correct species, the certified cfu and the typical characteristic biochemical reaction and morphology. Strains from cell culture collections like ATCC use sometimes several passages/subcultures until they are fit and show typical characteristic properties in the tests or on the medium. La norme ISO 16140-3 ne concerne que les laboratoires qui utilisent une méthode « alternative ». Ce n’est pas nécessaire si vous utilisez la méthode de référence (décrite par l’ISO).

Here ThermoFisher answer : We can quantify with the number of reads if prior validation of specific species in a defined matrix.

Merci Pierre !! Non mais pour de vrai, j'ai beaucoup d'espoir pour cette nouvelle solution. Comme toujours, il y a des points d'ameliorations à apporter, mais globalement ce nouveau système va aider énormément de laboratoire dans leur production d'eau peptonnée.

Hi Gael, as it´s mention on the User Guide, drops could remain into the groove of the head not affecting the filtration. Just remove them with a dry wipe and also get sure you are following the dry out step correctly. Hope this helps !

Hi Julie, not sure I do understand correctly your question but the funnel is the part to help to filtrate the product. The culture media plate, is where we put the membrane to let the bacterias grow. Is it answering your question ??

Analyser de l’eau n’a pas de risque pour l’opérateur (sauf si elle est extrememt contaminée). Donc le PSM servirait uniquement à éviter de contaminer votre eau. Donc je dirai que travailler dans un environnement propre devrait suffir.
