If you find the key to APOE,4, won't you solve the riddle of all Alzheimer's? You rarely talk about how APOE4 mishandles cholesterol and lets it accumulate in cells. Why not expand on this. My father's mother, sister and daughter all died of Alzheimer's disease which struck when they were around 64 or 65. Dead 10 years later. They all had one allele of Apoe4. My half sister was in her late 70s when she died and her mother died one week later at the age of 103. My father and his brother had early coronary artery disease and he drooped dead 3 months after his 3rd heart attack at 58. Most of these relatives had excellent diets, were physically active and had outgoing and vibrant personalities. Made no difference. I have had the most unhealthy living habits imaginable. I was addicted to sugar until a few months ago. I will skip most of my habits, By age 35, I started developing auto immune diseases as well as liver disease. MCTD, Lupus, schleroderma and 4 others. I started taking prednisone 25 years ago. Eventually I took it only every 4 to 5 days. Now I take it at 40 mgs every 3 weeks. I know it doesn't make any sense, but I will be 70 shortly, and I have no symptoms of Alzheimer's. I assumed I'd be dead long before this, but here I am and no Alzheimer's. Could it be related to the prednisone. I do have osteoporosis but every female in my family had it so it was inevitable anyway. I never get infections though my WBC are low. If you stop the inflammation any way you can, maybe you can prevent the development in the first place. I'd rather sacrifice my bones and keep my brain until the end. anyway.

Hi Rudy, I found you doing deep data mining on Alzheimer's for my final project in my medical assistant class which I got an A on btw, I also am an EMT and EKG tech, I've studied neurology so much in the past 2 years I could have a PhD in neurology, I have been doing this because it interests me and I have aquired covid induced Parkinson's, which is turning out to be a mimic form of parkinsons, there were several symptoms that indicate it is viral induced and I had them, the promonet symptom was a heavy vibration in the chest, I'm finding information every week on my condition, I believe it is curable based on tests are pointing at vagus nerve disfunction, which ultrasound seems to be helping, btw I was 5 when I Kennedy was shot too. Exactly the things downstream from the scene of the crime. Tau proteins happen when this disorder manifests. Quick onset is probably a gut bacteria gone rogue in the brain.

This was an excellent presentation on recent developments on ALZ disease mechanisms and hope for some treatments. My takeaway is that early detection of susceptibility (genetic or lifestyle) to amyloid plaques and/or neuroinflammation is possible along with possible early treatment to mitigate amyloid plaque buildup (which may occur for 10-30 years before becoming symptomatic)
