5:45 It seems like they lose weight with the isoleucine restriction initially but they all go up in weight over time. Maybe the effect of cutting this specific protein doesn’t last for a long period of time?

@@JesusChrist2000BC I was counting calories and weighing food on a scale. I was basically only eating carbs since I wasn’t eating protein and kept fat to a minimum. Maybe I was retaining more water with glycogen because of the extra carbs? Hard to know.

1:40 Protein restriction reduces body weight gain? It also increases energy expenditure? This is quite the opposite of all the common nutritional wisdom even given by doctors. Mainly that protein is the most satiating macronutrient, and that you need protein in order to maintain muscle mass.

It was observed since a very long time ago that high protein diet increase cancer and mortality. No one knew why at the time then we got the mainstream igf-1 hypotheses then reduced total protein in mice shown positive result and molecular studies of methionine and tryptophan then BCAAs shown some intriguing results then we tested in mice model then we got more data that these reduced these amino acids improve this and then whatever. I think the momentum isn’t there yet unlike the bad fat and bad carbs that recently get more attention so the evidence of these not-so-good amino acids have been piling for about a decade but it’s ofc very lacking compared to refined carbs and saturated fats. Prof. Dudley seem to make a big bet on this amino acids thing, and the data has so far supported this so i guess this might be the next big thing in nutrition sci in the next decade or maybe not because no one really want to reduce protein intake

@@Ziprextl Protein intake has also been shown to aid in muscle mass growth and retention. And muscle mass at later stages of life correlates with longevity. It’s seemingly 2 research areas that are at odds with each other.

@@DynamicUnreal I should stress that recommended protein intake which is about a chicken breast to most ppl, is enough to retain and grow muscle in most ppl. And that muscle retention and growth “particularly“ in older individuals depend much much much more on exercise than higher-than-recommended protein intake. The two main aa that have been identified to be very bad if you consumed a lot are isoleucine which is a BCAA and methionine, evidently in various mice models and very probably in human too due to recent studies that show some negation of benefits from vegan diets by adding them. This maybe why plant protein is better than animal protein, stimulating much less igf1 and insulin but i guess there also are effects from many absorbed dipeptides and tripeptides which we have almost no data about and in real food they come with a lot other things anyway so it’s even more complex. Whatever, take plant protein if you need extra protein although in fact, you should not take animal protein at all

Ultimately... it seems that survival of the species figures in to just about everything diet related. When food of all types is plentiful lifespan might be shorter, when only certain foods are available lifespan often increases. It appears we've been through countless of thousands of rough and good times in order to get our bodies to this point. What other secrets lie within?

Dear Professor, please excuse me if my comment merely shows my ignorance on the subject. The position of seniority in evolutionary age of the mitochondrial system along with it's being incorporated into and acceptance by so many different organisms tells a story of ubiquitous success. Its a well established, a settled in, a widely accepted, very old system that has stood up to all the evolutionary challenges so far. I acknowledge it's capable of getting sick and developing nasty mutations. Besides when such things might affect it, I can't see a reason to suspect that it plays a major role in aging, but who knows, it might. Genetics is definitely the right field to be in, but there's more than one tree to be barked up. Take for instance the bristlecone pine (Pinus longaeva) and a regular pine. The only thing that sets them apart from each other is the difference that exists in their genetics. Somewhere within that bulk of different code, are held the recipes / keys to its extraordinary longevity as compared with more regular pines. I suppose since you are looking at mitochondria, you might do well to compare the mitochondrial sequences so to capture the differences between that of Pinus longaeva and the other shorter lived pine species. That might provide interesting results much sooner than the ROS hypothesis. I'm not sure which if any may have been sequenced to date. If none have, then I guess it would be a good place to start trying to uncover new information. I hope you make lots more interesting discoveries. Wishing you the best of luck!

In my case, doing 16/8 TRE and eating a whole food plant based diet I am able to routinely elevate my ketones to clinical levels (0.5 mmol/L) plus or minus a bit for last few hours of fasting window. I also take this as a good marker for low insulin, which is desirable. If I restrict my calories for a few days, addition, similar to the Longo FMD, ketone levels will go up to 1.5 to 2.5 mmol/L which may be a good marker for increased autophagy, probably a good thing to do every month or two if you're older like me.

With so much discussion about rapamycin being the gold standard drug for aging, I wonder if modulating BCAA might be a good way to accomplish this as they are both accomplishing aging increases by reducing mTORC1 activity?

Perhaps, although rapamycin extends longevity in both males and females, whereas in the above work we only see life extension in the males. I'd also like to see a study in which both dietary methionine and isoleucine are selectively lowered to see if there are additive or synergistic effects.
