1. Anterior limiting membrane or anterior border layer 2. Stroma (Containing vessels,nerves,sphincter and dilator pupillae muscles, melanocytes,connective tissue fibres and extra cellular matrix) 3. Anterior pigmented epithelial layer which is continuation of retinal pigmented and then Ciliary body pigmented epithelial layer anteriorly 4. Posterior pigmented epithelial layer which is a continuation of non pigmented neurosensory layer of retina and non pigmented aqueous humour secreting non pigmented epithelial layer which comes to lie in the posterior most portion of iris and during embryonic development becomes pigmented and it's pigmentation gradually decreased towards the root of iris to become non pigmented epithelial layer of Ciliary body ! I read from Wolf's Anatomy yesterday and today in that it's given the posterior layer of optic cup comes to lie anteriorly and gets pigmented and becomes pigmented epithelial layer and the anterior epithelial layer of the optic cup that devlopes into neurosensory retina also into non epithelial layer of Ciliary body it comes to lie posteriorly in the region of iris due to arrangment optic cup is such that the non pigmented layer becomes posterior and pigmented layer becomes anterior in the region of iris while it's opposite in retina ! But you said here it's posterior layer of optic cup as pigmented layer of optic cup, in its anterior most region, but it's not so, it's not pigmented in Ciliary body even in retina but it becomes pigmented in the region of iris that which I see you said it by mistake, in that anterior most region of optic cup the posterior epithelial layer that's non pigmented becomes pigmented during embryonic development of iris and the anterior layer also becomes pigmented at the same time which is continuation of outer (that's is scleral side) pigmented epithelial layer which lies anterior or outer to non pigmented epithelial layer of Ciliary body which secretes aqueous humour and both the muscles sphincter and dilator pupillae are derived from mesenchymal cells which result from differentiation of the neural crest cells which again are differentiated products of neuroectoderm in the anterior most region of optic cup ! And Iris stroma and blood vessels develop from mesenchymal cells, which are developed from the second wave of neural crest cells and these neural crest cells are nothing but the differentiated products of neuroectoderm that lines the anterior most portion of optic cup region ! In short different layers of iris develop from 1. Posterior most pigmented epithelial cells from posterior non pigmented epithelial layer of Anterior most optic cup region which is a neuroectoderm which becomes pigmented during embryonic development and it's pigmentation decreases towards the root of the iris to become a non pigmented epithelial layer of Ciliary body lying on or coming in contact with the aqueous humour side 2. The anterior pigmented layer of epithelium devlopes from anterior layer of optic cup which is again a neuroectoderm 3. Stroma, blood vesels and sphincter and dilator pupillae muscles are derived from mesenchymal cells from second wave of neural crest cells which are differentiated cells derived from anterior most neuroectodermal epithelial layer of optic cup Development of iris simplified in three lines ! The blood supply from vessels of tunica vasculosa lentis and Long posterior ciliary artery formingna major arterial circle in the primary pupillary membrane region which starts getting reabsorbed from around 24 to 31 week of gestation age and this primary pupillary membrane and its vessels are derived from second wave of neural crest cells from this primary pupillary membrane that developes into the iris stroma and it's stromal structures including muscles and blood vessels ! In short the entire iris is developes from neuroectodermal cells ! Which differentiate into 1. Posterior non pigmented epithelial layer they become pigmented during embryonic development in the region of iris and the pigmentation is reduced towards iris root to become non pigmented epithelial layer of Ciliary body that comes in contact with aqueous humour, which secrete aqueous humour 2.Anterior pigmented epithelial layer of optic cup forms the anterior pigmented layer of iris which is continuous with the pigmented epithelial layer of the Ciliary body and retina 3. The anterior neuroectodermal layer of optic cup in its anterior most portion differentiates into neural crest cells causing the second wave of neural crest cells differentiating mesenchymal cells forming primary pupillary membrane the central or pupillary portion of which is resorbed before birth and para a papillary and peripheral portion of primary pupillary membrane forms iris stroma and blood vessels and sphincter and dilator pupillae muscles lying iris stroma ! For this simple thing, I have given my two hours to read iris development thoroughly from Wolfs anatomy, and today started reading iris anatomy from Wolf's, hope it helps you all ! I take so much joy in sharing my knowledge and skills !
