This seems wrong. If you measure from the top to the bottom of the sign cleat and make a line on wall then you would use this line and align the middle line of the cleat part on the wall with this line. The bottom of sign cleat would sit on middle of wall cleat.

You skipped over the part where you actually mount the letter on the wall. I'm guessing you screw the stud into the wall further than you need and turn it back to screw into the letter, with 2 nuts in the middle to tighten down on each end to set the spacing?
