I honestly never was worried about the trooper. He was just a stormtrooper, and Vader rarely hurt Stormtroopers. That, and he could read thoughts. He probably could read that the trooper was nervous and knew why he was leaning over(catching his breath).

A joke around bodily functions categorizes as 'slapstick comedy'. Well known jokes alike are fart and poop jokes. Mostly considered to be low brow comedy. But hey, no judgement here. Just the facts. The more you know... The fact that it's evil dictator DARTH VADER makes it even funnier because of his high status being undermined. We identify as and/or empathise with the stormtrooper, thus the tension of getting murdered if laughter is not contained. There are more layers but I won't bore you with it... In short: there's more to it than you'd think.
