If you see my other comment for additional details I think we are thinking in the same terms, although the Mr.Saikou's daughter part hit me out of left field I think that you could be right. Assuming Yandere's mother is a yandere like her then that would've meant she has a Senpai. For this situation to be true let's say Mr.Saikou was her Senpai but she failed in getting him but had been impregnated by him at some point. She settled for another man so her daughter would have a father figure in her life. At some point, Mr. Saikou's figure our after already having another child and another family, that he has a child with Yandere-chan's mother. At this point, he asks the headmaster of the school to watch out for his estranged daughter, know who she is. There are a lot of holes in this theory but it is a very interesting one. Alternatively, Mr. Saikou could be Yandere Chan's mother's brother and knows about his sister. He also could've her Info-Chan type person by in the day.

I have a theory about the headmaster deal. I think Mr Saikou was a former student at Yandere's school. Saikou told the headmaster that he suspect Ryoba (Yandere's mother) is the reason for the murders. The headmaster didn't believe it at first but he then saw Ryoba killing someone. She threatened him not to tell anyone. After the incident, Saikou and the headmaster had a deal. The deal was if Ryoba's daughter (Yandere) were to attend the school, Saikou's daughter won't attend the school. If Yandere is dead, the deal will be off and Megami would attend school. To put it in some commonsense, Megami is the last rival. If you kill any other rivals, Megami would be there to find Yandere and to put to the end of the killings. But if you haven't killed any rivals, Megami won't suspect Yandere as a murderer.

Here’s an idea that’s been bugging me for a while, what happens if you do this; 1. Kill Midori with a knife (wear gloves) 2. Dispose of the body and blood etc. but leave the knife 3.
