Despite the "lack of success" the video was still packed with lots of info for us. Have you tried agar with "antibiotic cocktail" - Streptomycin and Penicillin? Never fails. I too had one batch of wheat grain that persistently gave B.subtilis yet microwave rice packets inoculated from the same syringe were fine. Guess some stock is doomed with endospores. Keep up the excellent work.

Back again. I dug up that old wheat grain stock, soaked it overnight in 2% calcium hypochlorite and then Tyndallized it. Tyndallization is drain, boil for 10min, rinse and wait 24hrs before repeating this another two times. Three boils over 2 days before final bag and autoclave. Theory is endospores that escape sterilization hatch and go vegetative then they can be destroyed by heat. It's two weeks since innoculation with incubation at 28C. Firm mycellium ball has formed with no contamination. We shall see if it continues without problem. So endospores can be beat but try to use high quality food grade grain rather than animal feed. It is similar to semis in that they are grasslovers! Yes there is a name but I don't list the full names here on YT. Hint: They are named after a city in Florida starting with a T

Larva, moths and frass in your grains along with gnats, bring bacterial contams that wit seriously hamper mycelial growth. You hit it on the nail head. All that interferance introduces viroids as well. Like plants getting viroids, insects bring viroids to myco culture as well. Back to spore, new grains. Good sleuthing, Mate.

how long are you waiting to inoculate your jars? Do you inoculate within 24 hours Of PC cycle or do you wait longer? I feel as though that may be a variable that goes unnoticed. A lot of people say to inoculate asap after jars cool down. I think about uncle ben's packs, people have success with those and they sit on shelves for a long time.

There is a way around embedded contams it is a long tedious process but you could do water agar tank a clean culture off of that and then transfer it back to a plate

Your first plate from that one should have been water agar especially after you noticed The bacteria Yes the bacterial will still grow on the water agar but at the same time The mycelium will grow faster

With your grains I don't know how long you do your sterilization for but typically just avoid any contamination with mine I sterilize for 4 hours That's more along the line so The steaming process in the pressure cooker could actually get through the core of your jar not just the outer layer basically what's happening is your having contamination in the middle of your grains

I had the saying issue hidden contamination in my grams and I was pressure cooking for 2 hours with a 30 minute depressurizing. And I was having that issue so I moved mying up to a 4-hour cycle for pressure cooking to play it safe. Some people will say that you shouldn't do that because it will caramelize the sugars in the grain but pressure cookers at 15 psi Don't even reach the caramelization of sugar. Dave wombat on the Michael Gigi podcast was explaining that. And if you're having issues with your agar plates try water agar to clean it up.

I’ve tried water agar, but it’s never worked for me for embedded contams. The plate I was originally intending on just taking an aerial xfer but didn’t get around to it, then when I did I noticed the mycelium outgrowing the bacterial blotches. It’s very hard to clean an embedded contam because it doesn’t have blotches, the mycelium itself is contaminated at the core. The grains as stated in this video never had an issue prior to the moth infestation. All of the frass and other detritus from the moths introduce bad bacteria and viroids. I’d considered 4+ hour sterilization and maybe that would have solved it but I’d much rather get clean grains and be done with moths.

Ive been using high quality rye (organic, no signs of debris) and have been having inconsistent results as well with clean plates, 50% just wont fully colonize doesn't even look bacterial, I know they were properly hydrated by testing the grains with my teeth, I use modded lids with a 3nm filter patch, I PC for 150 mins Idk if its to long or what, but i'm going back to corn

As stated in the video the process is the same. It is most likely the dirty grains causing the bacteria as that is the main factor that has changed.
