Barr body from Buccal Cavity

Barr body Staining from Buccal Smear.
• Rinse the mouth with potable drinking water.
• Gently scrape the inside of cheek using a spoon.
• Discard the first scraping.
• Repeat the step and spread the scraping over a clean glass slide.
• Prepare a thin smear and air dry.
• Fix the smear with Carnoy’s Fixative for 30 minutes.
• Remove the slide from the coupling jar.
• Cover the smear with Giemsa Stain for 20 minutes.
• Rinse the slide under running water.
• Air dry the smear.
• Examine under oil immersion 100X lens of Compound microscope.
