Good job of covering the basics. I once was planning to go to Fresno State University, but life took a different twist. California State University was my choice instead. Glad things changed the way they did. Never thought I would endure what I did in Fresno, but God has a way of helping us see the sheer truth.

I think 0:49 has a mistake.It is not straight.

@@USA-ek1tl I think at 1:49 it is definitely not correct. The scan power is 4X. The low power is 10X (notice how they don't name it anything but 10X) and the high power is 40X. This is the conventional way of naming when these three lenses are on the microscope.

00:39 00:49 - Turn ocular lenses until #64 lines up with white line. 00:53 00:56 - Condenser lens must be just below the stage. 01:01 - Put objective lens on low power. Stage must be place at its highest position. 01:11 - Set up the light to medium or medium high range. 01:40 - Center specimen over condenser using the X/Y adjustement knobs. 01:46 01:58 - Make sure compound is centered in field of view on low power. 02:01 - Move to next highest objective lens (10 X) and refocus. 02:07 - Specimen nearly in focus; use fine-focus knob. 02:16 - Center specimen and move to highest power lens (40x). 02:40 - 1. The iris diaphragm is NOT used to adjust the light intensity, but to improve contrast and resolution. The microscope has a light intensity wheel on the base of the microscope for light intensity control.......

If the diaphragm is closed too much, you can lose resolution due to diffraction.

Hai~! First of all, the video is very good and the explanation given is also very easy to understand. However, there is one thing I want to ask (I hope the video maker or friends who see this comment can answer it ^^). So the question is, is there any particular reason that the ocular lenses needs to be set at 64? I thank you for this excellent video and also thank you for being willing to answer my questions :D

Before using a microscope , you need to wear a laboratory gloves to prevent dirt , dust and fingerprints in the specimen and to the microscope.

when using light microscope do you start the largest or smallest distance between the lens and specimen /why?

sorry if i'm late, but you should always start with the largest distance between the lens and the specimen. this is pretty much just because it is much easier to initially find the specimen with the lens, and to avoid making contact with the lens.

### Step-by-Step Process for Microscope Setup and Use 1. **Retrieve the Microscope** - Retrieve a microscope from the cabinet. - Carry the microscope with both hands: one hand on the arm and the other underneath the base. - If the microscope is too heavy, seek assistance. 2. **Set Up the Microscope** - Unwind the cord from the back of the microscope and plug it in. - Locate the on/off switch at the back of the microscope. 3. **Prepare the Ocular Lenses** - Adjust the ocular lenses until the number 64 aligns with the white line below the rotating part of the lens. 4. **Adjust the Condenser Lens** - Ensure that the condenser lens is positioned just below the stage. 5. **Select the Objective Lens** - Place the objective lens on low power (4X) and raise the stage to its highest position. - Adjust the light to a medium or medium-high range. 6. **Place the Slide on the Stage** - Open the clip on the stage and slide a microscope slide to the back of the stage, ensuring it is secure under the clips. 7. **Center the Specimen** - Use the knobs on the lower right side to center the specimen over the condenser lens. 8. **Begin Focusing on the Specimen** - Start with the low power objective lens (4X) with the stage in its highest position. - Look through the ocular lens and focus on the specimen. 9. **Increase Magnification** - Once centered, switch to the next highest objective lens (10X) and refocus using the fine-focus knob. - Again center the specimen before moving to the highest power lens (40X). 10. **Troubleshoot Focusing Issues** - If the slide doesn't move, ensure the right knobs are being used and that the slide is properly secured. - If you're struggling to find the specimen, reposition the stage to its highest position and start over at the lowest power. 11. **Observing Unstained Specimens** - Use the stage diaphragm to reduce light if observing unstained specimens. - Try to focus on bubbles or other easily observed objects under the cover slip. 12. **Clean the Lens If Necessary** - If a higher power lens makes the specimen appear blurry, clean the lens with lens paper and lens cleaner. 13. **Post-Use Procedures** - Remove the slide and rotate the lowest power lens into place. - Move the stage to a centered position and slide holder to the left. - Ensure the stage is clean and dry, turn off the light and power, and wrap the cord. - Carry the microscope with both hands back to the cabinet, ensuring the arm faces out and avoiding crowding.

Hello! There is a problem regarding my microscope... Basically earlier the coarse adjustment was working, and the microscope was going up and down accordingly... But now it's not working ( I think after I took the objective lens away from the slide by using the coarse adjustment, after sometime it got out of control) plz help in resolving the issue, it's a sincere request 🙏🙏🙏
