They needed natural resources like oil, aluminium, steel, coal etc. to run its industries, produce products, give jobs to citizens and food grains and crops to feed their population. Those resources were in the East. Barbarossa objectove was to get the Caucasus for the oil to continue the war effort. It was never about knocking down Soviet Union through Blitzkrieg which would be impossible anyway, considering the large landmass and population of the SU. The maximum timeline of Barbarossa was just 3 months with the fuel in their reserves.
