I just have one question that came to my mind before and it's driving me crazy: Are the quantitative value in the Wig file the read counts? If that is correct, shouldn't they be integer values? There's nothing in the wig format which says what has to go in the quantitation field. The value itself can be fractional so it's not requried to be a raw count. Most commonly you'll use some kind of normalised count (reads per million for example) so that you have some basic comparability between different samples. If it was only one sample you're looking at then you could use a raw count though.

Thank you so much this video. Your explanation on wig files was helpful. One question: what is the difference between "step" and "span". Do they have to be the same values? Thank you.

Span is equivalent to how large of a window the data value covers. The step is how far away the next window is from the previous one.
