That's all learned by being part of improv groups. The "games" played in them help with all aspects; teamwork, learning "yes, and..." (keeping scenes going), learning your style of character(s), sharpening smartass skills/responses, honing it as a craft, etc.)

for me the genius of these characters is pushing it just far enough that the other participant is confused but doesn't realise they're being worked, I guess its just me but when people are obviously in on the joke these scenes are not funny. Its overacted and the jokes become too cartoony to get a live reaction. he did it with ali g too and it really saw that character drift into mediocrity rather than just disappear a legend.
