Fragment b is always kept and Fragment a is thrown into the plasma, but there is one exception, it's for C2. So instead of keeping C2b , you keep C2a instead.

It's actually debatable, I observe, some of the older editions have it C4bC2b, even wikipedia says so. In my textbook it's C4bC2b.

In respect to the C4bC2b complex, for about 5 years or so in the recent past, immunology textbook writers wanted to make the process more uniform since C2a is the only large factor which remains associated with the cell surface and renamed it C2b to be more inline with C4b. So C4bC2b was published for a couple of years but it never really stuck as C4bC2a was already widely accepted and established.

I like how you started with the big picture first and then went into the nitty gritty details. My immunology prof doesn't do that enough, hence why I'm here. She will drill the process steps, but doesn't really get at the purpose of why these things happen. Thanks for this. :)

Hello AK lecture, according to my professor, C4bC2a is the C3 convertase that cleaves C3 to C3a and C3b. And also C4bC2aC3b is the C5 convertase and C3 convertase cannot cleave C5. Please keep on making video. thank you very much.

C4b2b is correct and it's the new form of formerly known C4b2a complex aka C3 convertase. Anyways, the name changed cause of some wrong nomenclature of complement proteins. The 'b' in these complexes stand for binding and 'a' stands for anaphylotoxins, so that should be it.

Hi I noticed that the C3 convertase should be C4bC2a ☺️ I looked it up in Immunology 7th Ed David Male page 90 fig. 4.4 ☺️

Actually, it was historically called C4bC2a. These days, scientists call it C4bC2b for the sake of uniformity. AK lectures is right.

Hi AK. Could you please check out this source: www.complement-genetics.uni-mainz.de/cp.htm I was reading Kuby's Immunology 6th edition (internation).. And the lecture notes supplied by university in UK also say it should be C4b2a.. really confused now..

Perhaps its an error in the book. Double check with your professor and let me know what he or she tells you.

Hey guys It all begins with the combination of antibody circulating to an antigen This complex attract C1 to bind here is the story. This complex will activate C4 and C2 to cleave into there( b) ones and( a )ones . Note any (a) constituent will initiate immune response . Any (b) constituent will bind to membrane, or to activate the others complements (only for c4 and c2) This complex after combination will give an activation for c3 to be (a) &(b) .any (a) will go to initiate immune response and for (b) will (b)ed on membrane. And so for C5

I'm so greatful for ur lectures I was abt to complain the same thing abt c3 convertase but I can see that it has already been done.I'm ur big fan... I appreciate ur metabolism videos on biochemistry.I'm hoping that immunology videos will help too. Pls prepare lectin pathway and also transplantation. If possible also prepare lectures on the other types of immunoglobins. I sincerely hope u would consider my pleas cos my exams is soon approaching... There are 2 groups of people. Those who call the larger active C2 fragment C3a and those who call it C2b. The larger active fragment of C2 was originally called C2a for some god awful reason and now there are people who want to switch around the names to go in line with the other complement proteins. Which makes sense, but it kinda just causes confusion.

My textbook describes the C3 convertase as C4b2a not C4b2b. it mentions when C2 is cleaved, fragment C2b leaves into the fluid phase and C2a joins with fragment C4b to form the C4b2a complex.

+tkanes17 You are correct the C3 convertase is composed of C4b and C2a, not C2b. I think this was a small mistake in the video as C2 is an exception to the general rule that the b fragment always is kept.

Great video! Very clear, thank you so much for posting. I do have one question though. When mentioning the C3 convertase you said that it is formed by a C4b and C2b molecule. However, in my text, as well as from my professor, I have been told that it is composed of a C4b and C2a, instead of C2b, molecule. I was wondering if there are any discrepancies in naming of the subpart of complement molecules after they are cleaved, or if this was just a small mistake? I apologize for the specificity and detail of my question, I just want to ensure I am understanding this process correctly. Thank you kindly, Kat

+Niiinjaaa Its not a mistake. As I said in a comment below this one, its currently known as C4bC2a but for a while people were pushing to change it to the C4bC2b to make it more logical. But people were simply not using it enough so I guess were stuck with C4bC2a for now.

C4b-C2a is the C3 convertase.

C3 convertase is made up of C4b and C2a. The portion of this video where c1 binds to IgM should result in C2aC4b complex instead of C4bC2b complex.
