Complex Matrices to Confident Multi-Omics Assays Enabled by AFA Technology

Advancements in the world of biotechnology have resulted a tremendous improvement in understanding of the genetic basis of diseases such as, Cancer, Alzheimer’s disease, and inherited disease. While the “Omics” world, in general, have undergone some significant evolution, the world of Genomics have moved ahead compared to others. The well optimized approaches in Genomics can ensure reliable extraction, isolation, purification of DNA and RNA from very complex matrices, ranging from fresh frozen tissues to FFPE, from cell lysates to whole blood. This Genomics workflow ultimately enables achievement of high-quality genomic data, regardless of sample complexity or user expertise. However, confirmation of a certain disease and its stage from Genomics perspective might not be enough and can benefit from confirmation obtained by monitoring the system from other Omics perspective. A multi-Omics based approach covering genomics, transcriptomics, epigenomics, proteomics, and metabolomics holds the key to highly confident data. While the precision medicine world continues to grow in size and importance, multi-Omics based assays for every system are still in its early days, and require both advanced technology for end-detection, and a consistent, comprehensive sample preparation and handling approach.
Adaptive Focused Acoustics® (AFA®) Technology has been the gold standard for DNA shearing in almost every Next Generation Sequencing (NGS) laboratory and is now gaining unprecedented popularity as the sample preparation technology for a host of Proteomics and protein analysis workflow. The results reported in this presentation will highlight increased reproducibility, reliability, robustness of the method in DNA shearing across different cell lines (S. Cerevisiae, Y. Lipolytica, E.Coli, L. Monocytogenes, Whole Blood, and S. Epidermidis) with very different characteristics. The workflows detailed in this presentation will also highlighting the benefits of a comprehensive workflow that results in high quality and high yield of DNA and RNA from Formalin Fixed Paraffin Embedded (FFPE) tissue samples. The data will show excellent %CV values along with high DV200 scores (for RNA) and high yield and amplifiability for DNA. The presentation will also highlight a one-plate workflow for protein extraction, isolation, digestion, and peptide purification from different biological matrices (fresh frozen tissues, tumor tissues, FFPE, organoids, cell lysates, plasma, etc.) In each example, in addition to a high number of protein ID, the AFA enabled workflows ensure high reproducibility.
Confident data results in accurate prediction - from understanding the behavior of a group of proteins to the Pharmacokinetic and Pharmacodynamic properties of a biotherapeutic monitored in plasma. AFA can enable robust, reliable and confident data for every analyte from any complex biological matrix.
