Apparently, Deadpool repeatedly forgetting his ammo bag became a running joke because the movie's relatively small budget was cut even more during production, so plans for more expensive gun-heavy action scenes had to be abandoned. It's ultimately why Wade only uses 12 bullets during the highway fight and relies mostly on his swords and hand-to-hand combat for the rest of the movie. But I'd say that it worked in the movie's favor because it made the fights more interesting than just a bunch of mindless gunfire.
