If Ast (req) < Ast (min) in that case what should we do ?

In that case you need to provide minimum reinforcement. If the minimum reinforcement also violates the maximum spacing criteria of main parallel reinforcement of slab, then you need to provide the reinforcements as per maximum spacing criteria of main reinforcement of slabs (As per IS456:2000).

Yes, it's quite simple, you just need to calculate the values of negative BM coefficients along the shorter and longer directions from Table 26 of IS 456, then calculate negative BMs along shorter and longer directions, then follow the steps to complete the design. As its interior panel, all four edged are continuous, you don't need to provide torsional reinforcement. The detailing part will be different. Just go through the detailing of the two-way continuous slab, you will understand.

What if 2 long edge discontinuous What is the value of Alpha X ?

if two long edges discontinuous, you will find the value of positive alpha x from table-26, pg no-91 of IS456, condition-6. Then calculate positive bending moment at mid span along x-direction, and negative BM along y direction and follow the steps to complete the design.
