Egg collection होने के बाद तय किया जाता है की patient को IVF , ICSI या IMSI करना पड़ेगा। IVF , ICSI या IMSI करने के बाद , इस fertilized egg and sperm जिसे zygote कहा जाता है, इसे incubator में रखते है ।Incubator बोहोत मत्वपूर्ण है embryo culture के लिए। IVF laboratory procedure में incubator का काम IVF में Embryo development करने के लिए एक स्थिर वातावरण प्रदान करना है।
IVF करने के बाद जब embryo को incubator में रखते है , तब embryo को closely monitor किया जाता है क्योकि इसका एक निर्धारित Timeline है जिसमे Mitotic division दिखाई देता है। ये Embryo development timelines बोहोत महत्वपूर्ण होता है जिससे पता चलता है की कितने embryos अच्छे quality के है या नहीं , या कितने embryos viable नहीं है।
120 -180 घंटे पे जांच की जाती है की ये Embryos , Blastocysts बने या नहीं। दिन के हिसाब से देखा जाए तो 3 रे दिन पे embryo division 8 cells की होती है इसकी जांच की जाती है और 5 वे दिन पे जांच करते है की ये blastocyst बना या नहीं। यही प्रकिरिया human body में भी होती है। इसलिए 5 वे दिन का blastocyst culture स्टेज बोहोत महत्वपूर्ण है।
Embryo Culture Media
बोहोत सारी कठिनाइया embryo को 1 Cells से लेके 100 Cells पोहोचने के लिए आ सकती है। Embryo को बढ़ने (grow ) केलिए Embryo Culture Media की जरुरत होती है। Embryo Culture Media बढ़ते embryo के लिए nutritional environment देता है है जिसे blastocyst phase में प्रवेश करता है।
Embryo Culture के लिए महत्वपूर्ण factors
१. Culture Media की स्टेबिलिटी और अच्छे क्वालिटी का Culture होना महत्वपूर्ण होता है।
२. High quality incubator का इस्तमाल बोहोत महत्वपूर्ण होता है।
३. Embryo development timelines भी बोहोत जरुरी होते है।
Embryo Processing
इस अंतिम प्रक्रिया में Embryo को freeze किया जाता है जिसको Embryo freezing कहा जाता है या Embryo Transfer किया जाता है। यह समझना बहुत महत्वपूर्ण है कि सभी Embryos , blastocyst phase तक नहीं पहुंचते हैं। बनाए गए सभी Embryo में से, 30-40% blastocyst phase तक पहुंचते हैं।
