Thank you for the informative video but on 23:30, how total number of runs can be 12? should not it be 64 from 2^6?

It's a fractional factorial design so the number of experiments is decided beforehand. The number of runs you decide determines your resolution and each resolution (III, IV, VI, Full) has a certain risk to it (check out 34:31). You are essentially saving time but sacrificing the amount of knowledge you get from the experiment. For a full factorial design, it would take 64 runs and take significantly more time.
