The endothelium is a single cell layer lining the arteries and veins that is in constant contact with the blood and it functions to aid blood clotting for hemostasis and to prevent or slow unneeded clotting.

what about the Glycocalyx Structure that sits on the Endothelial cells? I think that this is far more important for if it is functioning properly you have no trouble, but if it is damaged then you have leakage. Your Thoughts?

Yes, the glycocalyx provides protection of the endothelium from damage and surface over which the blood cells can freely flow. Heparan which activates antithrombin III and nitric oxide synthase which makes NO for vasodilation are important elements of the glycocalyx.
