I think it's a combination of funny scripted ideas with actual bloopers. There are some that were definitely natural. Like the "everypony" one and Luna's classic "furpessional" moment. You can hear, I think it's Tara that said "that was really good..." and you can hear the cast laughing in the background. The "glue glow" Pinkie one I think was also real and Andrea was goofing around and improvising. You can hear Rebecca (Sunset) is genuinely cracking up.
