Is it possible to change a schedule with excel without the sync button ? Can we export the excel file, edit it, then manually import it back ? It's more convenient than the live sync button.

The error you are experiencing might be due to a conflict with another Revit plugin that also uses ‘DocumentFormat.OpenXml’ for reading, manipulating, and writing Excel (.xlsx, .xlsm) files. You might want to temporarily uninstall the other plugin and test again.
