what does the Detune do for the beat? does that mess with auto tune at all?

No. He's just kinda stating that you could tune it back up 50% to get the sound of the original draft. I think lol. Either way a detuned beat that is already exported should not have an effect on autotune, your daw will automatically assign what key the track is in, and then would find that and manually adjust autotune settings to match the key.
