You see 3 bars (usually) above a piece: HP, Mana and Cooldown of the skill.

That's a very good question! Since it's not about the golem buff but another unit, I would say yes, it does buff Kira if the golem is melee. Tho I've never tested it. I'll try it out and come back with the correct answer. :)

I would go with Doom. If the opponent has her as the most expensive unit, it can be probably silenced with a Doom. If not, then a Grimtouch chain + Doom combo can do the trick. If you don't have the opportunity to go for these, I would try with some area silencers, like Lava Shaman or Storm Shaman, but these are less effective imo. :)
