Gene Therapy

When we looked at some areas of biotechnology earlier in the series, we briefly touched on gene therapy, without saying much about what it is. Now we are ready to dive into this topic and see what it is all about. If a person has a defective gene, what can be done about this? Is there a way to modify the genes in someone's cells? How exactly would that work? Should we be doing this at all? We will answer all of these questions and more in this video!

Cool video. I’m part of gene therapy study right now. I had severe hemophilia- after the therapy I now only have mild hemophilia. The therapy was attached to a vector virus.

I think engineered meganucleases or TALENS have a lot of promise for genome targeting. Still, I feel that most treatments are going to be restricted to diseases that can be treated by transplant of ex vivo transfected cells.

mr dave how electron absorb and EMIT photons. i watched many of your videos about modern physics and others and also googled but couldn't find the answer. i know that electrons emits photons when it changes its energy level in atom but how electron emit/produce photons? electrons are made of quarks so do they change while emitting photons ???

electrons are not made of quarks, they themselves are fundamental particles. as to emitting photons i'm not sure how that works

@Nandan Ramesh A very simplistic way of thinking about it is smth like this: since electron is moving, it produces a magnetic filed, or in other words emitting EM waves (radiation); on a different energy level the nature of this motion changes, therefore the field (or waves) produced changes as well. This difference is represented by releasing a quant of EM radiation - photon. On more technical level, there aren't really particles, all of them, including photons and electrons, are exaltations in respective quantum fields. Electron field of the universe constantly interacts with electromagnetic field; so when there is a change in electron's energy level, this energy excites EM field, producing what we register as a photon. There's a great lecture on quantum fields by David Tong at The Royal Institution channel, definitely check it it out if you're interested in these things. Or at least there you'll find a much better articulated answer =)

I have nf1 which causes my body to develop random tumors. How promicing would it be for this? Like there's no way my existing tumors would just dissapear right? New ones develop periodically and the existing ones just slowly get bigger. Hope this made sense Proffesor Dave.

If they're actively suppressed in the healthy body then genetic therapy would suppress even existing tumors.

Hi Prof, few years back the gene therapy was only limited to the autosomal recessive diseases.Since we could only insert the desired normal allele into the genome. In your video you mentioned that the retrovirus could inactivate the mutated gene and allele , this is quite a latest information ( maybe the gene therapy is not limited anymore , it can be applied to autosomal dominant diseases?) Do you have any sources of article for this and what kind of vector that researcher use in that case ?

Hmm, I don't think I said that it could inactivate the mutant gene, nor do I think that's the case. But you don't need to, as long as the healthy protein is being expressed, it doesn't matter that the mutant protein is also present since it is nonfunctional.

So, the new gene cells are inserted, and they start to replicate. The new gene cells...rupture(?) and the virus with the RNA sequence infect the older cells, mutating them? Is that sort of close? I apoligize if I'm sounding like an ignoramus, this kind of science (along with a LOT of other diciplines) fascinate me. I'm trying to distill it down to something I can wrap my head around.

(Sorry for the awful grammar) If we dont make it a race thing amd just focus on objectivly justifyable properties like for exanple eyesight.
