Nice explanation taking you thru the steps of algae grow. Just have a few question Sunlight = your grow lights , Water = obvious water in glass flask , Oxygen = oxygen you pump into the glass flask with co2 , ... But what are you using for Chlorophyll = ??? in your lab and Glucose = ??? in your lab. Just curious as i am trying to figure out how to make algae on a small home scale for just hobby projects. Thanks for any reply.

Most algae use the steroids, sunlight, and the CO2 to produce glucose and chlorophyll so, they actually don't need either added to their environment! Hope this helps!

@@happyphace163 sucrose at 3% is used to accelerate growth, it supplies glucose even before photosynthesis starts the cycle. It is standard in plant tissue culture.

Clumped up algae is a good sign. It means that the algae is getting enough light, and that they are most likely alive. You can even see it happen in the video here, when they pan across some of the younger cultures. If something still isn't right, for some reason, then the Co2 should help stop it, but it may still occur. Hope this helped! :)

If crude oil is the best carbon sink, and since we can already make algae into crude oil, can we make oil and store it? Making the crude oil would take a lot of energy, but say we used solar energy.Could we sequester enough C02 out of the atmosphere to slow down climate change? How much crude oil do we need to make?

Biocrude is the way to go. In addition to dealing with CO2 atmosphere problems, it is also a great alternative to the geopolitical nightmare that has evolved because of the petrodollar economy. Biocrude and a biodollar economic model allow for greatly improved carbon capture, more energy self-reliance, etc.. Unfortunately, the EPA has obstructive laws that criminalize use of waste produced CO2. Algae Industry Magazine is free online.

michele antinozzi Purchase from a culture collection like UTEX, perform dilution spread plates. Isolate colonies that do not have biological contaminants in them, restreak these colonies onto a fresh agar plate to build up enough biomass to inoculate a liquid culture, and finally add streak to culture flask at a minimum of 0.2 OD730. Do all of this in an absolutely sterile hood. Do not do this at a bench, as seen in this video. I can promise that if you do, every single one of your cultures will have bacteria growing in it.

If you're looking to start a culture to feed your crags, I'd start a Nannochloropsis culture. It can tolerate a large scale of salinity.

Ritesh Bhagea I am not involved in their production, but if you want to know how much to use, in general, you want the air bubbles to pass a greater distance so that sufficient gas exchange occurs before the air bubble exits the flask. If enough starter culture is present to start with a good density of cells, I'd recommend as much liquid before your set-up starts to bubble onto the stopper.

I get you sterilize your flasks and instruments with your autoclave but if one had a pressure cooker for a hobby project the should be ok to. My issue is i not sure what you add to your water and what water source your using to be exact on all the steps shown. Thats really the only issues to actually go and try this some day!

@Gabriel Atienza Using tap water? Can't do that, might contain trace amounts of chloride/chlorine. I'm about to try it/ experiment with it, think as long as I provide some nutrient, enough sunlight and decent water source it should work good
