Mamizou only appears in Extra stage and is an extra stage boss. You cannot play it on easy or any other difficulties since Extra is considered a separate difficulty. And even ignoring that Mamizou had really tough cards, for me the hardest one was her survival spell. And that's also not counting the Extra stage itself.
