I'm not in the mycelium-business but in my field of work hygiene plays an important role. we usually have nested rooms where air particle count is reduced step by step. This technique could be useful for you if you either put your labs inside a larger tent that prefilters the air (probably overkill, I admit) or add some kind of pre-filter to your hepa filters so the large particles get taken out beforehand. If you have enough sucction you might even be able to use a cyclone system which is very cost effective during operation.

The resilience you have is great. Even when anticipating contamination I know it still hits hard. Keep on pushing man !

You want to buy some Bentonite. I once read about an experiment, in which some guys sprayed a hospital wall with Bentonite mixed with water. Another wall was NOT sprayed with bentonite. Over two years they monitored the bacteria levels on each wall. The wall they had sprayed with Bentonite was bacteria free - after two years!!! Maybe you can use this and spray it onto the insides of your tunnels, the shelving and also rinse out your bags with it. Check it is food grade, however.

I think the biggest issue is that your Lab is right next to the front door so all of the spores from outside can be blown or carried inside whenever entering/exiting. There seriously needs to be a means of isolating the area where you do your lab work and the entryway of the shed.

I hate to hear this guys!! That being said, I am do glad you show what's going on and give an in-depth explanation of how you are going about finding the source of the problems, what you're doing to remedy it and what your plan is moving forward. This is such great information for anyone either starting out or wanting to better their business. So, sorry for the setback but keep up the great work! We love your channel and can't wait to see how things move along!

A quick way to sterilize rooms, jars, bags, and all things ( except for live Mycelium grain spawn) is to use UVC light.

the great thing about the hardwood pellets is that they are pre pasturized. you may get away with less sterilizing time therefore somewhat quicker cool down time. every time i mess with my bags when they are still hot i get problems

I load my unicorn bags in the 55 gallon steamer with the folded side down run it for 18 hours and unplug it. I throw on some welding gloves and unload after 10-20 min cool down. I am using 50/50 soy hulls and sawdust. 7lbs of substrate per bag. I have to watch when I am loading that I don't dump a bag out but after the sterilization run the bags are pretty much vacuum sealed when I pull them out.

Just an idea, build air locks around doors. As you walk in, the outside air is exchanging so the air cleaners are being bypassed. Air locks will prevent air exchange when entering/exiting.

If your quick and sloppy infront of your flow hood air locks aren't the answer. A chip fab lab is cool when you can operate it in the correct way.

@@pacificnorthwestfreshmushr930 Airlock space must be more than 1 step wide. Open outside door, close outside door, walk two or three steps to open inside door to enter lab. It was just a thought. I implemented such an airlock design on the front of my 20' geodesic dome. I had 12' airlock where I stored wood. The inner and outside doors were never open at the same time which prevented air exchange.

I tend to use Unicorn bags with T filters for grain spawn runs, then transfer under clean laminar flow to master's mix using A filter bags for a bit more breathability, pore size is still too small for most foreign fungi spores to enter the sterile bag. Most contamination issues is due to the grain to bulk substrate transfer, still air in a clean room with the laminar flow HEPA filter running for an hour or two should completely clean the room of spores and dust. Otherwise it could be the sterilisation process, best to use heat under pressure e.g. pressure cooker/autoclave, or a barrel autoclave for large scale that runs for 24 hours. You can also get the built in injection port Unicorn bags which reduce contamination risk and can even be inoculated with LC out of laminar flow.

I work in the biotechnology field. We grow virus, cells, and bacterial cultures that ultimately are processed into antigens and vaccines. In our class A labs we alternate between disinfectants weekly and monthly. Try using Spor-klenz once a week in order to kill those hard to kill spore forming bacteria and other unwanted mold spores.

I’m my home lab I will put on a 24hr cycle of ozone that gets up to 4000ppm ozone, then I switch on a 265nm UV flood light, that adds a second layer of sterilisation but also neutralises the ozone. After that I leave an agar dish out for 10 minutes uncovered to survey, literally not even one CFU detected after a fresh cycle so far

Hi again, Brian! Word of advice: All you have to do is pop the lids on your drums off and leave the bags in there for a 24 hour period. Then they've cooled off enough to where you can safely move them into your lab and leave them for another 24 hours to further cool. Then inoculate. It should not be necessary to seal the bags before you sterilize them. Keep up the great work man!

WHAT THE FUNGUS Noob question, but why can't you run cool water through the sterilizer for a few hours prior to handling?
@johngrady1755
@johngrady1755 6 жыл бұрын
I believe the bags are not sealed, which would be a problem. Secondly, they have a filter patch which would allow water intake. Besides those two things, you have to use water, which is more money/work than simply waiting 24 hrs.

I immediately started adding up the cost of loss in the truck bed. I feel your pain. Why don't yall use liquid culture to inoc bags so no sealing cutting inoc. and resealing? Is it for less steps or so no puncture in bag to have to worry about contamination point? Costs more too i guess.

Damn. Lost the whole farm lol. Contams happen but that's just insane. If you didn't start out in a sab and jumped infront a flow hood from the get go you did yourself a major disservice. Its a good video but it doesn't tell the whole tale. Forest fires aren't the issue. I personally believe every grower needs to start off in an sab. Your movements are constructed in an sab. I think this is an example of what cutting corners can look like. Its a business but the mushrooms don't know that and your flow hood doesn't know that either. Do an sab video. Doing the wrong things for a long time over and over again results in the same things. Everything down to the movement to where tools are positioned infront of a flow hood or sab is what I've learned. Jars and sabs should be a way every commercial grower starts. I don't think I've heard you speak much on still air boxes or what goes on inside one. Those critical steps determine the outcome of your farm and how it will operate in the future. I wouldn't advise to anyone looking to do this to take out a massive loan to go big or go home.
