Female Jonathan is always a cool concept, but I don’t see why Dio would treat her really different from regular Jonathan. Maybe more respect since she would be a woman but Dio’s bi. If he wasn’t attracted to Jonathan he wouldn’t likely be attracted fem!Jonathan. (Still a cool concept, I really like it. Not saying ya’ll are wrong, it’s an au after all, I’m just saying what would probably happen knowing Dio. The only person he ever truly cared about was his mother after all)

He would treat her differently but not because of attraction or love, but because Dio's ambitions, Victorian society and gender roles would come in play and it would have him approach a female Jonathan differently. Dio was all about getting that Joestar fortune and one way to do it would be marrying female Jonathan and seeing how George admired Dio I would see him being OK with him marrying his daughter. Female Jonathan being a woman in Victorian England would have less rights and less of a say in anything and Dio wouldn't so much have to exert his superiority and establish dominance like he did with male Jonathan because he would expect to have that over her already (and Dio would underestimate her like he did with male Jonathan). After he becomes a vampire however most of that would not matter but depending how their relationship plays out (did they just grew up together? where they fiancees? husband and wife? Wid they have a child?) their dynamic would be affected by it. As for caring about Jonathan. Judging by what Dio says to Jonathan after he's surprised that Jonathan survived Dio's minions and by what Dio said to him in the ship, I think Dio did end up caring about Jonathan in the extend he is able to care about someone else. Of course all of that didn't really do much good to Jonathan since it's Dio's positive feelings towards him and ambitions that ended up with Dio stealing his body.

Personally I think he would treat her differently. You see OG Dio (no mask) hated the good nature of the Joestars and looked down on them but he wanted their fortune. His competition for that fortune was Jojo. Who was a man in old Britain. He was passive, very loving and kid and forgiving. Dio did not like that. But if Jojo was a girl, those traits in his personality would be more acceptable to Dio because usually those attributes in olden days was viewed as a good wife material or what not. Which leads me to the next thing, Dio wouldn’t have competition to inherit the fortune. As a woman, Jojo wouldn’t inherit shit if we are going by that times standards. The likelihood of Dio getting paired with Jojo seems very plausible as we know how Jojo’s dad was. I mean he viewed Dio as amazing, he was a boy and to keep the fortune in his bloodline he just had to give his daughters hand in marriage. Like to me it makes sense because Dio was just bad natured and ambitious. This would’ve been an easier more convenient path in my eyes. So yeah :v that’s just my take. You might be absolutely right but to me, it just isn’t about attraction but that the power dynamic and status and situation changes if Jojo was a woman.

And Dio could still be an isolating prick that JoJo can punch real good. But it depends on what their relationship can turn into. I can also see a yandere route happening if Dio does care for JoJo. The vampire thing can also be something he struggles to choose or not. It’s very interesting.

He's already top tier waifu material. And he should love Jojo for who she is and support her to get stronk and be her own person. I don't know if Dio would do that for Jojo. I just want Jojo to be happy for once and for her to be loved and protected. And it would be fun to throw Dio challenges to see if he can be a better man or if he goes off the rails. A tragic ending would be that he does love Jojo and she loves Dio back, but he still chooses to become a vampire.
