Very common to not wear gloves in a microbiology lab. Reason is, experience has shown that techs are less likely to wash their hands if bacteria or chemicals end up on them. Most bacteria are basically harmless on a healthy skin surface, as long as you wash them off right away. If you have cuts or similar wounds on your hands though, then you should definitely wear gloves.

Depends on the species of course. Most are not pathogens and the normal skin flora will keep many in check. Using bugs to fight other bugs.

My mixed unknown project is coming up. First step is to streak 2 TSA plates in a way to isolate the 2 bacterial colonies. Then I have to prepare a bacterial smear of each colony to perform the gram stain of each. Next I have to create pure cultures of the bacteria of each colony. The question I have is this. Would the colonies appear purely isolated on the TSA plate in order for me to do the next 2 steps?

How identification and characterization of bacteria can be done 3. If I have a single bacterial strain- and I have to culture it. Was there a reason you didn't cool the loop as much on the second plate? On the first plate you double tapped the loop and paused a couple seconds. On the second plate you single tapped and immediately smeared it. It just caught my eye so I asked. Cool video though.

For no particular scientific(?) reason, really. I used to intern at a microbiology lab and I do that sometimes. The double taps and pause for the first quadrant was probably because the loop was a bit too hot.

You're wasting so much time. Petri dishes are not held like that in the open. Spores fall on the agar.

That's because the tube will not be used to cultivate microorganisms, apparently. So, as they will be sterilized and discarded, it is not necessary to flame the neck of the tube. Although you're right if the culture in the tube will be kept.
