Basically Jenson is talking about his photo op session with the fans and how some of them wear make up. Since during the photo ops he generally is hugging the fans if they request it, the make up might rub off on his shirt or what have you. So Daniela gave him a reversible jacket so once the photos are done he can flip it to the other side and not have to walk around with make up smears. She then tells him to take off the jacket, presumably so the make up on the jacket won't rub off on his black shirt and force him to have to change out of it.

She is organizes the annual convention in Rome, Italy, JIBCON, over the last ten years she has become good friends with the cast. they all love doing the Rome convention. Jensen is very good friends with her, always makes sure she gets a big Thank You. She bought him this coat as a gift and then she, for a laugh, got in the question line and asked him why he was wearing a different coat from the photo ops, giving him a chance to show off the reversable jacket, and then get him to remove it so he wouldn't be too hot on stage. They are like brother and sister at this point it seems, such a good friendship.
