Not quite. Misaki was always like that to Usui. Kanou didn't like the fact that Misaki was the student body rep. I believe the context was that Kanou didn't like Misaki imposing strict rules; and that he might have a slight phobia to women. That's why he put a hypnosis on her such that if she fell asleep, she would be entirely under his control. This was why Misaki stayed tried to stay awake the entire time (well that, and she's very stubborn and hot blooded).
