@@DIYBiotech a lot of room for unethical misuse too, especially exposure/implementation without consumer awareness; the potential misuses violates inherent medical autonomy. I hope you have a conscience and forsight to the ways people/government/corporations might use your work/efforts. In a sense it is genomic assault, if areas of the body are affected without permission/against our will. We saw how vocid triggered DNA changes in organs through CRISPR/mnra tech after people across the planet were forced to receive "treatment".

As someone wanting to pursue biology research and someone who has recently found and now love and almost depend on the benefits of THC and psilocybin, I've become increasingly worried about being perceived as a drug addict and also becoming one, but seeing you succeed and finishing something this big while also releasing public videos with interest in these compounds gives me hope

Maybe, but ever since that day I am an academic weapon as well Don’t let other people’s perception of you change your life But be careful with shrooms, they are good once every often to ground and guide you, too many and you may lose your focus and purpose in life Same goes for LSD, I’ve lost a couple of years on a side quest They weren’t wasted, but for sure did delay my life quest for a bit as I was learning more spiritual stuff

I watched several DIY Biotech vids (he made a bunch a couple years ago). I do not remember noticing any indication the video creator enjoys consuming THC or psilocybin as much as you do. He might, but I doubt it.

Is there a good strain of yeast which has been engineered to break down waste carbohydrates without generating undesirable volatiles? Or could you setup an air pump into a bioreactor and use a carbon filter to scrub the smell?

As a software engineer, I've started hearing about how badly people like me are needed in fields like bioinformatics and I used to REALLY like science back in high school, so I've been thinking about going something more in this direction. I am completely self-taught and dropped out of school my sophomore year, but I've been thinking about going back (for NOT a comp sci degree lol) and I'm curious what kind of degrees you would suggest I look into if I want to work with yeast!! Seeing the broad spectrum of naturally occurring psychoactive substances makes me wonder what other kind of natural compounds can be manufactured with yeast

There's a HUGE need for coding types in biotech right now. I'm sure you could slide into a biotech position with just comp sci background. If you're looking for a master's or PhD in genetic engineering type things, look into biochemical engineering, bioengineering, or biological engineering programs. Definitely look to see what professors are researching before applying. My lab is chemical engineering, but we only do biology.

Extraordinary video! One question: you’re scanning ALL potential [gene knockout locations? CRISPR guides?], for optimal gene placement location/efficiency, then manually evolving them to further suit the task by making the yeast report production r8 for you??? you’re essentially just plugging the output activation from the biosensor (programmatically) into growth factors or other genes? This is really cool!

Some questions about the last chapter. It seems like one of the knockouts produces more terpenes simply because it can grow faster whereas the other one optimized some terpene synthesis pathway. Did you make a strain with both knockouts? Also, did you use barcodes when making your library or did you sequence the entire genome of your increased fitness mutants to find the KO. Congrats on passing and great video!

Great questions! The two best knockouts grow the same on regular media, they only grow different when there are high concentration of terpene added to the media. Both knockouts would be a good next step! Yes, each sample of our genome-wide screens are barcoded so we know which flask the guide counts came from. We only have to sequence the gRNA on the plasmid, not the whole genome. Another reason why CRISPR screens are great!

But you clearly know your stuff. 😺 I was really curious though, near the end when you're talking about the two knockout strains vs the base, did you create a strain that had BOTH of those genes knocked out? Really curious to see whether production would go up more or whether they would interfere with each other. Or is that like, hard to do for some reason?

Great question. One of the professors on my committee asked the same thing! No I didn't try that, but yes, there is a good chance it would improve titers even more. But, these genes also serve other functions, so the more knockouts you make, the more likely the overall health of the cell decreases

Tried Bradford but got huge differences in protein content comparing to kjeldahl... don't know why that happens...what did you use? Would be great to get your opinion on that 😊

Can you do work by adding "feeding energy by electricity" to Yeast from electric-powered (electric feeding) bacterias? Its seems can be interesting for industry "precision fermentation microbiological protein production" for example, for all Yeast-based industry

Two things here: 1. you can make carbon sources with electricity and CO2 (artificial photosynthesis) 2. you can provide energy to cells in the form of electricity But energy and carbon source are two separate things, just to be clear

Do you capture the gas released by the yeast and study its composition to see what the yeast is in excess of or lacking in their environment? Kinda like measuring the chemical expressed when your gut bacteria die or whatever to send chemical signals to you.

I know that Krasnoyarsk Institute of Biophysics lab "Bioluminescence of sea" work for Bayer - they searched genes of bioluminescence for Bayer's did sensors like you do for automatic cellar drags experiments (100 000 tests per a day line, thay said). How it can be helpful for your work?

Hello, congrats! Your research is very interesting. I am also doing a phd in Biotech and I wanted to ask: Do you got training during the first few weeks of a laboratory class? I take laboratory classes where the professors don't train us whatsoever and delegate all the work on us. To me it seems they're just lazy. Is that the way it is in phd programs or not?
