Robert Koch discovered M. tuberculosis in the year 1882 and the same year Paul Ehrlich reported the acid fast nature of M. tuberculosis. M. tuberculosis is not stained by Gram staining because of its cell wall nature. All bacterial cell wall is made up of Peptidoglycon but tubercle bacilli cell wall high content of lipids, fatty acids and mycolic acid. Later this method also modified by Ziehl and Neelsen
