No, the max velocity is 10 in your case. But Km kicks in at half of the maximum velocity of enzyme activity (5).

If you can't estimate it by yourself with your data set (which shouldn't be that difficult to begin with), then you could use a Lineweaver-Burk plot (or another linearisation) and calculate Km and vmax off of that.

Hello, thanks for the video it was very helpfull, my question is i have a very low [s] and Vmax and excell didnot want to give me the Km value. i have tried the same values as the video and i get the exact answer but when i try my own data i dont get any thing at all. (my data is as low as to the power of -06) may be excell doesnt work on such low values? (am using mM as a unit for S and umol/min for Vmax).

You need to set your solver to look for the minumum and not the maximum.

There are some really great tutorials on editing out the mouth smacking noises on your video. If you search for "how do I stop disgusting saliva sounds when recording," you should find a bunch.
