Western Blotting is a common technique employed in most life sciences labs. An increasing number of journal editors and reviewers are requesting that the associated data is acquired using a camera-based imaging system that assures much greater accuracy and dynamic range in quantification of band densities than film. However, most of the software tools that come with gel and blot imaging systems lack the densitometric analysis tools to perform accurate analysis of the bands with normalization to a loading control protein or total lane. Bio-Rad recently launched the Chemidoc MP Gel and Blot Imaging System to perform precise and sensitive imaging of gels and blots coupled with the ImageLab software tool which simplifies data acquisition for chemiluminescent, colorimetric and fluorescent samples. The software also features integrated data analysis tools to not only perform sophisticated densitometry but also a normalization tool to assure that the resulting band densities are not only quantified accurately but also normalized to the reference protein or the total protein lane. This video tutorial demonstrates how ImageLab is used to both acquire and analyze data from a multifluorescent blot with normalization.
