Novel Strand-Specific Approach for AAV Genome Titer and Genome Integrity Determination Using DNA Hybridization and Biolayer Interferometry
Abstract:
The presence of complete viral vector genome, that constitutes e.g. promoter, enhancer, gene of interest, polyA tail, for efficacy of the therapeutic is becoming clear. This necessitates genome integrity determination.
Currently long-read sequencing and multiplex dPCR are the most commonly used methods for genome integrity evaluation. New approaches that do not involve PCR amplification, such as biolayer interferometry (BLI) shows promise of high accuracy, fast run time and high repeatability and precision. Additionally, the technology provides a strand specific quantitative information that could not be obtained by any other technique.
We have evaluated the applicability of the assay for genome titer quantification and vector genome integrity. The results have demonstrated appropriate repeatability and accuracy, providing strand specific quantitative information. The strand specific titer capability lends itself well to genome integrity determination of self complementary AAVs (scAAV).
