what are the dimensions of knuckle part? and how to design it in the nx ?

Yes; if only loads or constraints change, then you re-run the Nastran solution, “clone” the Durability solution, and re-solve. However, if there is a design change that requires re-meshing, you would “save-as” the finite element model and re-solve both Nastran and Durability solutions.
