can I Keep the co2 screwed down or once punctured needs replaced after the trip on the water? How long would you recommend keeping it screwed in before I should replace?

for a kayaker, this would be handy with a standard flotation jacket, but I wonder since it inflates in front, would it push you on your back and get in the way or you getting back on the kayak? Anybody out there use this on a fishing kayak and can report back?

Yes you can use the oral tube to manually inflate the vest.

There's literally a five year old video on rearming and repacking the M-16 from the manufacturer. It even shows up as a suggested video.
