I think the problem you have with the white petal that you painted red and still showed white around the edges, its the type of brush you are using. You have 100% opacity, but the brush effect is diffused, it gives diffused edges. Maybe use the pen tool, or another with a hard edge. I cant be more specific because I havent used that software.
