Kindly go slow and make neater diagrams rather than overlapping on the slides. Sometimes we lose the track of diagram u explain manually. Most importantly please improve your English. But overall your explanation is surely amazing, I have a presentation to make on microscopy, I have referred to your videos whilst preparing for it.

Hello sir! Have a small question. You said if no specimen is present all you see is a dark background. But the cone of light coming from the condenser through the glass slide has to get refracted somewhere right? Doesn't a part of it pass through the objective lens? If so then you should see somewhat a bright background instead of dark background..

this interpretation of 90 degree phase shift is wrong, the phase change inside the specimen is not 90 degrees but a very small value, the phase change of the background light is not to get a destructive interference, but to get a largest slope with respect to the phase information of the specimen. if a destructive interference is achieved at the focus plane, then the contrast only comes from the scattering effect of the specimen but not from the phase information of it. in order to get phase information, the phase relation between the scattered and the unscattered light has to be 90 degree, because here the derivative of the cosine function has the largest value, and the intensity is very sensitive to the phase change caused by the specimen. when it is 180 degree as it is interpreted in the video as the destructive interference, the derivative of the sine function is 0, and the intensity doesn't change with the phase change caused by the specimen, therefore no phase information is resolved. I'm quite sad to see this wrong understanding of the phase contrast microscopy is held by most of the biologists and widely distributed.
